Epidermal Lymphocytes

Epidermal lymphocytes are immune cells residing within or immediately beneath the epidermis, where they provide local surveillance against tissue damage, infection, and abnormal cell growth. Most are T lymphocytes that recognize antigenic signals through their receptors and respond by releasing cytokines or engaging directly with stressed or transformed cells; communication with keratinocytes and other immune cells helps shape the local response. In cancer research, these cells offer insight into early tumor immunosurveillance, immune evasion, and the inflammatory microenvironment of skin cancers. Their distribution, activation state, and interactions may inform biomarker development and strategies for improving immunotherapies targeting cutaneous malignancies.

Dermatitis herpetiformis (DH) is a chronic inflammatory condition characterized by an autoimmune reaction between IgA and epidermal transglutaminase (eTG). DH develops in a very small portion of gluten-sensitive and/or celiac patients. The results of this study indicate that DH can also develop in a rhesus monkey host with symptoms of idiopatic dermatitis.

Lymphocytes are the major players in adaptive immune responses. Here, we present a lymphocyte purification protocol to determine the physiological functions of the desired molecules in lymphocyte activation in vitro and in vivo. The described experimental procedures are suitable for comparing functional capacities between control and genetically modified lymphocytes.
