Nickel Grid Preparation

Nickel Grid Preparation is the process of cleaning, coating, and loading nickel electron-microscopy grids with biological specimens for stable imaging. These grids provide a conductive support for ultrathin tissue sections, cellular fractions, or immunolabeled samples, while nickel’s chemical compatibility can suit specimens requiring an alternative to copper. During preparation, a support film is secured to the grid, the specimen is mounted and contrasted or immunogold-labeled, and excess liquid is removed before electron microscopy. In cancer research, this workflow helps visualize tumor-cell ultrastructure, organelles, virus-host interactions, and molecular markers at high resolution, supporting studies of disease mechanisms and treatment response.

2011

Visualizing protein samples by negative stain electron microscopy (EM) has become a popular structural analysis method. It is useful for quantitative structural analysis, such as calculating a 3D reconstruction of the molecules being studied, and also for qualitative examination of the quality of protein preparations. In this article we present detailed protocols for preparing the EM grids, staining the sample and visualizing the sample in an electron microscope. Novice users can follow these...
