Peritoneal Cavity Model

A peritoneal cavity model is an experimental system that recreates the abdominal serosal environment to study how cancer cells survive, disseminate, and establish tumors within the peritoneum. In common animal models, researchers introduce tumor cells or tumor tissue into the cavity, where interactions with mesothelial surfaces, extracellular matrix, immune cells, and peritoneal fluid influence implantation and metastatic growth. This approach helps characterize peritoneal carcinomatosis, tumor burden, ascites, and treatment responses in cancers such as ovarian, colorectal, and gastric cancer. By linking disease progression to a defined anatomical site, the model supports investigation of tumor biology and evaluation of localized or systemic therapies.

The peritoneal cavity in mammals contains different immune cell populations crucial for innate immune responses. An efficient isolation method is required for biochemical and functional analyses of these cells. Here we provide a comprehensive method for the isolation of peritoneal cavity cells in the mouse.

This video demonstrates a peritoneal lavage method for isolating mature connective tissue-type mast cells from the peritoneal cavity of adult mice. Upon isolating a mixture of primary immune cells from the peritoneal cavity, selective proliferation of mast cells is performed under appropriate culture conditions.
