Tumor Cell Expansion

Tumor cell expansion is the controlled increase of tumor cell numbers under laboratory conditions, enabling researchers to obtain sufficient material for cancer studies. Cells are maintained in culture vessels with suitable nutrients and growth-supporting conditions, then periodically passaged to limit overcrowding and sustain proliferation while preserving relevant cellular characteristics. Expanded tumor cells support experiments on cancer biology, drug response, treatment resistance, and interactions with the tumor microenvironment. When carefully monitored for viability, morphology, genetic stability, and contamination, this process provides a reproducible foundation for comparing tumor behavior and developing more representative models for preclinical research.

An efficient protocol for the ex vivo expansion of tumor-reactive T cells from tumor-draining lymph nodes or other secondary lymphoid tissues of tumor-bearing hosts is described. This protocol selectively expands tumor-specific T cells for use in adoptive immunotherapy of breast cancer.

Ex Vivo Expansion of Natural Killer Cells from Peripheral Blood Mononuclear Cells

This video demonstrates a technique for the expansion of natural killer (NK) cells directly from peripheral blood mononuclear cells (PBMCs) using a feeder cell line. PBMCs are co-cultured with genetically modified lymphoblastoid feeder cells expressing membrane-bound interleukin-21 (mIL-21), and the culture is supplemented with the cytokines interleukin-2 (IL-2) and interleukin-15 (IL-15). Together, this cocktail of ligands leads to the prolonged expansion of NK cells with high purity.
