Tumor Chip

A tumor chip is a microengineered, three-dimensional model that recreates key features of a tumor and its surrounding tissue in a controlled laboratory device, supporting more physiologically relevant cancer research than conventional cultures. Microfluidic channels maintain living tumor cells with extracellular matrix and, when included, stromal or immune cells; controlled fluid flow creates nutrient, oxygen, and drug gradients that influence tumor growth and treatment response. Tumor chips help researchers study cancer progression, invasion, cell interactions, and therapeutic resistance while testing anticancer drugs under tunable conditions. By connecting engineered models with patient-derived cells, this approach may improve personalized treatment selection and reduce reliance on animal studies.

This video demonstrates chip-based digital PCR — a variation of the digital PCR technique that is useful in detecting rare transcript variants. The PCR reaction is partitioned into the chambers of a nanofluidic chip, each of which acts as an independent reaction. The detection of fluorescence signals from the chambers with amplified targets confirms the presence of rare transcript variants in the sample.

A lensless on-chip fluorescent microscopy platform is demonstrated that can image fluorescent objects over an ultra-wide field-of-view of e.g., >0.6-8 cm2 with <4μm resolution using a compressive sampling based decoding algorithm. Such a compact and wide-field fluorescent on-chip imaging modality could be valuable for high-throughput cytometry, rare-cell research and microarray-analysis.
