Zebrafish Xenotransplantation

Zebrafish xenotransplantation is a cancer research technique in which tumor cells from humans or other organisms are introduced into zebrafish embryos or larvae to study tumor behavior in a living vertebrate. Researchers commonly label transplanted cells with fluorescent markers and place them into accessible sites, while the larvae’s small size, optical transparency, and immature adaptive immune system support real-time imaging of tumor growth, cell migration, invasion, and interactions with blood vessels. This model enables rapid assessment of metastasis and anticancer drug responses, helping investigators compare treatments and examine cancer mechanisms in vivo. Its speed and scalability also support personalized oncology and early-stage therapeutic screening.

A Standardized Pig to Macaque Heterotopic Heart Xenotransplantation Model

This study presents a pre-clinical heterotopic abdominal heart xenotransplantation model using α-Gal knockout (GTKO) pigs and macaques. The model incorporates optimized recipient selection, vascular anastomosis techniques, and perioperative management strategies. We describe a protocol for xenotransplantation into the yolk of transparent zebrafish embryos that is optimized by a simple, rapid staging method. Post-injection analyses include survival and assessing the disease burden of xenotransplanted cells by flow cytometry.

In this protocol, porcine specific primers were designed, plasmids-containing porcine specific DNA fragments were constructed, and standard curves for quantitation were established. Using species-specific primers, cpsDNA was quantified by qPCR in pig-to-mouse cell transplantation models and pig-to-monkey artery patch transplantation models.
