Clinical-grade Production

Clinical-grade production is the controlled manufacture of chemical substances that meet predefined standards for identity, purity, quality, and consistency in clinical research or medical use. In chemistry, it applies validated processes, documented procedures, qualified equipment, and testing under appropriate quality systems, often following Good Manufacturing Practice principles. Analytical methods monitor raw materials, intermediates, and final products, while process controls and batch records support traceability and reproducibility. These practices enable reliable scale-up from laboratory synthesis to clinical supply, reduce contamination and variability risks, and provide the quality evidence needed for regulatory review and safe evaluation of new therapies.

Here we present a novel clinical grade isolation and culture method for kidney Perivascular Stromal Cells (kPSCs) based on whole organ perfusion with digestive enzymes and NG2-cell enrichment. With this method, it is possible to acquire sufficient cell numbers for cellular therapy.

Protocol for propagation of dissociated high grade glioma surgical specimens in serum-free neurosphere medium to select for cells with cancer stem cell phenotype. For specimens that fail to grow as neurospheres, an alternative protocol is suggested. A paraffin embedding technique for maintaining the 3D neurosphere architecture for immunocytochemistry is described.

Here, we present a protocol to achieve the large-scale manufacturing of adherent cells through a fully closed system based on GMP-grade dissolvable microcarriers. The cultivation of human mesenchymal stem cells, HEK293T cells, and Vero cells was validated and met both quantity demands and quality criteria for the cell and gene therapy industry.
