Column Overload

Column overload is a chromatographic condition in which the amount or concentration of sample introduced exceeds a column’s capacity to retain and separate compounds effectively. When excessive solute occupies the stationary phase, retention becomes nonlinear, causing peak fronting, band broadening, reduced resolution, and inaccurate quantification. In analytical chemistry, recognizing column overload helps researchers optimize injection volume, sample concentration, flow rate, and stationary-phase selection. Preventing overload produces sharper, more symmetrical peaks and improves the reliability of compound identification, purity assessment, and quantitative analysis in liquid and gas chromatography.
