Planar Chromatography

Planar chromatography is a separation technique in which compounds migrate across a flat stationary phase, making it useful for identifying, comparing, and assessing chemical mixtures. In techniques such as thin-layer chromatography and paper chromatography, a solvent serves as the mobile phase and moves through the stationary layer by capillary action; compounds travel at different rates because of their differing affinities for the stationary and mobile phases. After development, separated spots can be visualized and compared using retention factors. In chemistry, planar chromatography supports reaction monitoring, mixture identification, purity assessment, and rapid qualitative analysis, often providing an efficient preliminary method before instrumental characterization.

Supported planar bilayers are powerful tools that can be used to model the molecular interactions in an immunological synapse. Here, we show methods for anchoring cell adhesion proteins known to modulate synapse formation to the upper leaflet of the lipid bilyer and visualize synapse formation using TIRF microscopy.
