Single Beam Spectrophotometer

A single-beam spectrophotometer is an optical instrument that measures how much light a sample absorbs or transmits at selected wavelengths, making it useful for identifying and quantifying chemical substances. It passes light from a source through a wavelength selector and then through the sample to a detector; the instrument compares the transmitted intensity with a separate blank or reference measurement and calculates absorbance, commonly using the Beer-Lambert law. In chemistry, single-beam spectrophotometers support concentration measurements, compound characterization, reaction monitoring, and analyses of solutions in research and teaching laboratories. Their straightforward design also provides an accessible introduction to instrumental analytical methods.

Microvolume samples are quantified by a spectrophotometer system that uses natural surface tension to retain samples without the use of cuvettes or capillaries. The dynamic range of protein concentrations and speed by which they can be measured are greatly increased with this method.
