Tetra-n-butylammonium Fluoride

Tetra-n-butylammonium fluoride (TBAF) is a soluble organic fluoride salt used as a fluoride source and reagent in synthetic chemistry. In solution, its tetra-n-butylammonium cation separates from fluoride, allowing the small, nucleophilic anion to attack electrophilic centers; most notably, fluoride cleaves silicon–oxygen bonds in silyl ethers to release alcohols and form stable silicon–fluorine bonds. Chemists use TBAF for desilylation, fluoride-mediated transformations, and activation of silicon-based reagents in organic synthesis. Its reactivity depends on solvent, concentration, temperature, and water content, since commercial preparations may be anhydrous or hydrated, making handling and reaction control important.

In this protocol, we demonstrate how to breed Astyanax mexicanus adults, raise the larvae, and perform whole-mount immunohistochemistry on post-larval fish to compare the phenotypes of surface and cave morphotypes.

18F-Labeling of Radiotracers Functionalized with a Silicon Fluoride Acceptor (SiFA) for Positron Emission Tomography

The synthesis of fluorine-18 (18F) labeled radiopharmaceuticals for positron emission tomography typically requires months of experience. When incorporated into a radiotracer, the silicon-fluoride acceptor (SiFA) motif enables a simple 18F-labeling protocol that is independent of costly equipment and preparatory training, while reducing precursor quantity needed and utilizing milder reaction conditions.
