Airway Remodeling

Airway remodeling is the long-term structural and functional alteration of the bronchial wall that can develop after persistent respiratory disease, particularly asthma, and contribute to fixed airflow limitation. Repeated inflammation and repair can promote smooth muscle hypertrophy, subepithelial fibrosis, epithelial injury, and enlargement of mucus-producing glands, narrowing the airway and increasing its reactivity. In clinical research, airway remodeling helps explain changes in lung function, symptom persistence, and disease severity, while informing assessment of anti-inflammatory and disease-modifying treatments. Understanding these changes may support earlier intervention and therapies aimed at limiting irreversible airway obstruction.

Nucleosome Remodeling

2020

Nucleosomes are the basic units of chromatin compaction. Each nucleosome consists of the DNA bound tightly around a histone core, which makes the DNA inaccessible to DNA binding proteins such as DNA polymerase and RNA polymerase. Hence, the fundamental problem is to ensure access to DNA when appropriate, despite the compact and protective chromatin structure. Nucleosome remodeling complex Eukaryotic cells have specialized enzymes called ATP-dependent nucleosome remodeling enzymes. These enzymes...

The goal of this protocol is to provide automated methods to quantify chronic lung pathologies in a murine model of COPD. The protocol includes exposing mice to cigarette smoke (CS), measuring pulmonary function, inflating the lungs, and using morphometry methods to measure emphysema and small airway remodeling in mice.

COVID-19 / Coronavirus Outbreak: Protecting The Airway - Endotracheal Intubation

2020

In pandemic times, medical staff is becoming a key resource in fighting the infection. To achieve the best medical care, relevant techniques and procedures have to be taught to medical staff to reduce the risk of infection. COVID patients often need mechanical ventilation due to progredient respiratory insufficiency, so an endotracheal intubation becomes a critical procedure in managing these patients. This procedure has an increased risk of infection due to aerosol formation and working with...

A Model of Cardiac Remodeling Through Constriction of the Abdominal Aorta in Rats

A rat model of abdominal aortic constriction that induces cardiac hypertrophy and remodeling is described. An efficient, highly-reproducible, and minimally-invasive method is used to provide a simple yet useful platform for research in myocardial hypertrophy and dysfunction.

A Method to Target and Isolate Airway-innervating Sensory Neurons in Mice

Organ specific sensory neurons are difficult to identify. Fast Blue tracing is used to identify nodose neurons innervating the airways for cell sorting. Sorted nodose neurons are used to extract high quality ribonucleic acid (RNA) for sequencing. Using this protocol, gene expression of airway specific neurons is determined.
