Biomarker Receptor Occupancy

Biomarker receptor occupancy is the measurement of how many target receptors are bound by a drug or other ligand in a biological system. The approach typically uses a labeled tracer, binding assay, or imaging probe to distinguish available receptors from those occupied by the test compound, with occupancy influenced by dose, exposure, affinity, and competition at the target site. In clinical research, receptor occupancy biomarkers help confirm target engagement, guide dose selection, and relate pharmacokinetics to pharmacodynamic effects. They can also support evaluation of drug efficacy, safety, and treatment response, particularly for therapies acting on receptors in blood cells, tissues, or the central nervous system.

Determining the fungal diversity within an environment is a method utilized in occupational health studies to identify health hazards. This protocol describes DNA extraction from occupational air samples for amplification and sequencing of fungal ITS regions. This approach detects many fungal species that can be overlooked by traditional assessment methods.

We describe a rat model of post traumatic stress disorder (PTSD) that reveals the persistent alterations in neuroendocrine function and the delayed long-term, exaggerated fear response, characteristic of PTSD patients. The animal model and methods described here are useful for correlating biomarkers in brain nuclei, which are mechanistic but cannot be measured in patients, with biomarkers in peripheral white blood cells, which can.

Here, the techniques used to transfect synthetic microRNAs into host erythrocytes and isolate all RNAs from P. falciparum are described.
