Drug Protein Interaction

Drug-protein interaction describes the binding of a drug to a protein, an interaction that influences how the drug is distributed, transported, metabolized, and acts in the body. Binding occurs through forces such as hydrogen bonding, electrostatic attraction, hydrophobic interactions, or covalent attachment, and can alter protein conformation, drug affinity, or the fraction of drug available to enter tissues and reach its target. In clinical pharmacology, characterizing these interactions helps explain drug efficacy, duration of action, toxicity, and variability among patients. It also supports the prediction of drug-drug interactions, optimization of dosing, and development of safer, more effective therapies.

This video demonstrates a method to study protein-drug interactions using QCM biosensor-based biopanning of the T7 phage-displayed peptide library. The biopanning of phages displaying a specific peptide on the QCM biosensor allows specific isolation of high-affinity peptides, followed by amino acid sequencing, which helps to identify specific interaction sites on the target protein.

Presented here are two antibody-based protein-protein interaction research techniques: immunofluorescence and immunoprecipitation.
