Embryo Lysate

Embryo lysate is a preparation made by disrupting embryonic tissue to release its cellular contents, providing a complex molecular snapshot of the developing organism. The process yields a mixture of soluble proteins, signaling molecules, metabolites, lipids, and nucleic acids whose composition depends on embryonic stage and preparation conditions. In developmental biology, these components can be analyzed to characterize changing molecular environments or applied to cultured cells to examine effects on signaling, proliferation, and differentiation. Embryo lysate therefore supports studies of embryonic regulation, cell-cell communication, and tissue development while offering a biologically complex material for cell culture and regenerative research.

Isolation and Purification of Kinesin from Drosophila Embryos

This is a protocol to isolate active full length Kinesin from Drosophila embryos for single-molecule biophysical studies. We show how to collect embryos, make the embryo lysate, and then polymerize microtubules (MTs). Kinesin is purified by immobilizing it on the MTs, spinning down the Kinesin-MT complexes, and then releasing the kinesin from the MTs via ATP addition.
