Ethanol Fixation

Ethanol fixation is a sample-preservation technique that stabilizes cells and tissues for microscopic and molecular analysis, making it valuable for examining developmental changes. Ethanol removes water from the specimen and precipitates proteins, limiting enzymatic degradation while preserving cellular and tissue architecture; fixation conditions such as concentration and exposure time influence morphology and downstream staining. In developmental biology, researchers use ethanol-fixed embryos, larvae, and reproductive tissues to document anatomical patterning, perform histological staining, and detect selected molecular markers. The method is rapid and compatible with many laboratory workflows, although dehydration can cause shrinkage or alter some structures and can reduce the quality of certain assays.

Microdialysis of Ethanol During Operant Ethanol Self-administration and Ethanol Determination by Gas Chromatography

A method to determine the time course of ethanol concentration in the brains of rats during operant ethanol self-administration is described. Gas chromatography with flame ionization detection is used to quantify ethanol in the dialysate samples, because it has the sensitivity required for the small volumes that are generated.

Fluorescence in-situ hybridization (FISH) is often required in combination with histopathology and molecular diagnostics for selection of therapy in personalized medicine. A novel non-cross-linking, formalin-free tissue fixative that allows high quality morphologic, molecular and FISH analyses from the same specimen by addition of a post-fixation step before FISH is presented.
