Human Corneal Fibroblasts

Human corneal fibroblasts are connective-tissue cells found in the corneal stroma, where they help build and maintain the extracellular matrix required for corneal strength and transparency. In response to injury or biochemical signals such as transforming growth factor beta, relatively quiescent keratocytes can become activated fibroblasts and, in some conditions, contractile myofibroblasts that produce and reorganize collagen and other matrix components. Studying these cells helps developmental biologists investigate corneal stromal formation, cell-matrix interactions, wound healing, and scar formation. Their use in culture and tissue-engineering models also supports research into corneal repair, disease mechanisms, and regenerative therapies.

Carcinoma-associated fibroblasts (CAFs) rich in myofibroblasts present within the tumour stroma, play a major role in driving tumour progression. We developed a coimplantation tumour xengraft model for experimentally generating CAFs from human mammary fibroblasts. The protocol describes how to establish CAF myofibroblasts that acquire an ability to promote tumourigenesis.

Here, a protocol to isolate and establish primary fibroblast/myofibroblast (MF) cultures from frozen gastric, small intestinal, and colonic tissue-yielding cells with a MF phenotype-is presented. These cells express CD90, α-SMA and vimentin. MFs can be used for a variety of functional assays including enzymatic activity and cytokine production.

Establishment of Proliferative Tetraploid Cells from Nontransformed Human Fibroblasts

Although proliferative polyploid cells are necessary to analyze chromosomal instability of polyploid cells, creating such cells from nontransformed human cells is not easy. The present report describes relatively simple procedures to establish proliferative tetraploid cells free of a diploid population from normal human fibroblasts.
