Spatiotemporal Manipulation

Spatiotemporal manipulation is the controlled alteration of a biological event across space and time, a framework that helps reveal how developing organisms organize cells and tissues. In developmental biology, researchers regulate the location, timing, duration, or intensity of signaling pathways and gene activity, then measure how these changes affect cell fate, migration, proliferation, and tissue patterning. Localized perturbations and time-controlled interventions can distinguish whether a signal acts directly, depends on its duration, or produces different outcomes in distinct regions. This approach clarifies mechanisms of morphogenesis and developmental disorders while supporting more precise experimental models of tissue formation.

Typical microtubule inhibitors, used widely in basic and applied research, have far-reaching effects on cells. Recently, photostatins emerged as a class of photoswitchable microtubule inhibitors, capable of instantaneous, reversible, spatiotemporally precise manipulation of microtubules. This step-by-step protocol details the application of photostatins in a 3D live preimplantation mouse embryo.

2014

Pneumonia is one of the top ten leading causes of death in the U.S. Osteopathic manipulative techniques (OMT) may be utilized as an adjunctive therapy in the treatment of pneumonia to enhance biomechanical and immune function; and ultimately, to reduce hospital stay, duration of antibiotics, incidence of respiratory failure, and mortality. In this video-article, we will review randomized controlled studies on the use of OMT in pneumonia patients, as well as demonstrate specific hands-on...

Recently available video recording and spatiotemporal mapping (STmap) techniques make it possible to visualize and quantify both propagating and mixing patterns of intestinal motility. The goal of this protocol is to explain the generation and analysis of STmaps using the GastroIntestinal Motility Monitoring (GIMM) system.

This article describes a video imaging technique and high-resolution spatiotemporal mapping to identify changes in the neural regulation of colonic motility in adult mice. Subtle effects on gastrointestinal (GI) function can be detected using this approach in isolated tissue preparations to advance our understanding of GI disease.
