Zeeman Slower

A Zeeman slower is a device that reduces the speed of an atomic beam, enabling atoms to be captured for laser cooling and precision experiments. It combines a counterpropagating, frequency-detuned laser with a spatially varying magnetic field, whose Zeeman shift compensates the changing Doppler shift as atoms slow and keeps them resonant with the light. This radiation-pressure force repeatedly scatters photons and decelerates the atoms while the magnetic-field profile maintains efficient interaction along the device. Zeeman slowers are important components of cold-atom systems, including magneto-optical traps, atomic clocks, quantum sensors, and experiments that require controlled low-velocity atomic samples.

We present a parametric driving method to cool an ultracold Fermi gas in a crossed-beam optical dipole trap. This method selectively removes high-energy atoms from the trap by periodically modulating the trap depth with frequencies that are resonant with the anharmonic components of the trapping potential.

We describe photobleaching methods including Fluorescence Recovery After Photobleaching (FRAP) and Fluorescence Loss In Photobleaching (FLIP) to monitor chromatin protein dynamics in embryonic stem (ES) cells. Chromatin protein dynamics, which is considered to be one of the means to study chromatin plasticity, is enhanced in pluripotent cells.

Inositol pyrophosphates play an important role in human pathologies such cancer, diabetes and obesity; however, the exact mechanism of action is a matter of dispute. The lack of commercially available inositol pyrophosphates renders detailed studies problematic. Here we describe a simple protocol to produce and isolate milligrams of inositol pyrophosphates.

Assessing Cerebral Autoregulation via Oscillatory Lower Body Negative Pressure and Projection Pursuit Regression

2014

Cerebral perfusion is maintained across a range of pressures via cerebral autoregulation. However, characterizing autoregulation requires prominent pressure fluctuations at regulated frequencies. The described protocol will show how oscillatory lower body negative pressure can generate pressure fluctuations to provide data for projection pursuit regression for quantification of the autoregulatory curve.
