Environmental Gradients

Environmental gradients are systematic changes in physical, chemical, or biological conditions across space or time, such as shifts in temperature, moisture, salinity, light, or nutrient availability. These gradients arise when environmental factors vary along landscapes, depth profiles, or seasonal cycles, creating conditions that influence organismal physiology, species distributions, community composition, and ecosystem processes. Researchers use gradient-based observations and experiments to compare responses across conditions, identify ecological limits, and assess how organisms adapt to environmental change. In environmental science, this framework supports habitat assessment, biodiversity monitoring, conservation planning, and predictions of ecosystem responses to climate change and other disturbances.

Density Gradient Ultracentrifugation

2023

Density gradient ultracentrifugation is a common technique used to isolate and purify biomolecules and cell structures. This technique exploits the fact that, in suspension, particles that are more dense than the solvent will sediment, while those that are less dense will float. A high-speed ultracentrifuge is used to accelerate this process in order to separate biomolecules within a density gradient, which can be established by layering liquids of decreasing density in a centrifuge tube. The...

Environmental Enrichment for Rodents

2026

All animal procedures described here must be conducted in accordance with institutional animal ethics guidelines and approved by IACUC. All procedures must follow the principles of the 3Rs—Replacement, Reduction, and Refinement—and must be performed by trained personnel. Environmental enrichment involves objects and practices that encourage natural behaviors, improve psychological well-being, and promote better welfare in laboratory rodents. Environmental enrichment should be tailored to the...

Source: Rustad, M., et.al. Synthesis of Infectious Bacteriophages in an E. coli-based Cell-free Expression System. J. Vis. Exp. (2017)This video demonstrates the purification of bacteriophages from a suspension containing host bacterial debris using a sucrose density gradient. It shows how the density gradient separates phages by buoyant density, enabling recovery of a visible band for resuspension and further analysis.

Aseptic Technique in Environmental Science

2023

Source: Laboratories of Dr. Ian Pepper and Dr. Charles Gerba - The University of Arizona Demonstrating Author: Luisa Ikner Aseptic technique is a fundamental skill widely practiced in the field of environmental microbiology that requires a balance of mindfulness and practice in the laboratory. Proper use of this technique reduces the likelihood of bacterial or fungal contamination of reagents, culture media, and environmental samples. Aseptic technique is also vital to ensure data integrity and...
