Fungal Its Region Sequencing

Fungal ITS region sequencing is a DNA-based method for identifying fungi and characterizing fungal communities, especially in environmental samples where many species cannot be cultured. It targets the internal transcribed spacer (ITS) region of ribosomal DNA, amplifying this variable genetic marker with PCR before sequencing and comparing the resulting sequences with reference databases. The approach supports species identification, taxonomic classification, and metabarcoding of soil, water, plants, and other habitats. By revealing fungal diversity and community composition, ITS sequencing helps researchers investigate ecosystem function, environmental change, plant-microbe interactions, and the distribution of potentially beneficial or pathogenic fungi.

We describe methods for live-cell video microscopy of Candida albicans phagocytosis by macrophages. These methods enable stage-specific analysis of macrophage migration, recognition, engulfment and phagosome maturation and reveal novel aspects of phagocytosis.
