Bibac-gw Vector

The BiBAC-gw vector is a specialized binary bacterial artificial chromosome (BAC) cloning vector designed to maintain and deliver large genomic DNA fragments for genetic analysis. It combines BAC-based replication and maintenance in bacteria with Gateway recombination sites, allowing an insert to be transferred efficiently from an entry clone into the destination vector. T-DNA border sequences support delivery of the cloned region into plant cells through Agrobacterium-mediated transformation, where the DNA can integrate into the plant genome. This capacity enables functional studies of complex loci, including regulatory regions and gene clusters that are difficult to analyze with conventional plasmids.

We describe the production strategy of integrase-deficient lentiviral vectors (IDLVs) as vehicles for delivering CRISPR/Cas9 to cells. With an ability to mediate quick and robust gene editing in cells, IDLVs present a safer and equally effective vector platform for gene delivery compared to integrase-competent vectors.
