Modified Mrna Delivery

Modified mRNA delivery is a method for introducing engineered messenger RNA into cells so they can produce a selected protein without altering genomic DNA. Chemical changes to nucleosides can improve mRNA stability and reduce innate immune sensing, while carriers such as lipid nanoparticles protect the molecule and promote cellular uptake; once released into the cytoplasm, the mRNA is translated and then naturally degraded. In genetics, this approach supports transient protein expression for vaccination, protein replacement, and delivery of genome-editing components. Its tunable design and nonintegrating activity make it valuable for studying gene function and developing therapeutic strategies.

2025

This video demonstrates the process of transfecting primary macrophages with modified mRNA encoding green fluorescent protein. These modified mRNAs, designed to reduce immunogenicity and improve its stability, ensure the successful transfection and subsequent expression of green fluorescent proteins, which is confirmed through fluorescence microscopy.

This protocol presents Rayleigh breakup as a mild process for aerosolizing non-viral mRNA vectors. Regional deposition for aerosolized mRNA vectors is evaluated by the Alberta Idealized Nasal Inlet (AINI) model. pre-mRNA Processing

2019

In eukaryotic cells, transcripts made by RNA polymerase are modified and processed before exiting the nucleus. Unprocessed RNA is called precursor mRNA or pre-mRNA to distinguish it from mature mRNA. Once about 20-40 ribonucleotides have been joined together by RNA polymerase, a group of enzymes adds a “cap” to the 5’ end of the growing transcript. In this process, a 5’ phosphate is replaced by modified guanosine that has a methyl group attached to it (7-Methyl guanosine). This 5’ cap helps the...
