Non-coding Variants

Non-coding variants are DNA sequence changes that occur outside protein-coding regions, yet they can strongly influence gene activity and genetic traits. They may alter regulatory elements such as promoters, enhancers, silencers, or splice-control sequences, changing transcription-factor binding, chromatin accessibility, RNA processing, or the timing and level of gene expression without modifying a protein’s amino acid sequence. In genetics, studying these variants helps explain findings from genome-wide association studies and reveals mechanisms underlying disease susceptibility, development, and variation among individuals. Integrating sequence analysis with functional assays and gene-expression data can identify which non-coding changes have biological effects and guide research into diagnosis and therapeutic targets.
