Papillary Fibroblasts

Papillary fibroblasts are connective-tissue cells located in the superficial papillary dermis, where they help maintain the skin’s extracellular matrix and support communication between skin cells. Their behavior is shaped by gene expression and signaling pathways that regulate collagen production, matrix remodeling, cell growth, and responses to injury. In genetics research, these cells provide a model for studying how inherited or acquired changes influence skin structure, wound healing, aging, and disease. Comparing papillary fibroblasts with fibroblasts from deeper dermal layers can also reveal how location-specific genetic programs produce distinct cellular functions.

This manuscript describes a FACS-based protocol for isolation of papillary and reticular fibroblasts from human skin. It circumvents in vitro culture which was inevitable with the commonly used isolation protocol via explant cultures. The emanating fibroblast subsets are functionally distinct and display differential gene expression and localization within the dermis.

Carcinoma-associated fibroblasts (CAFs) rich in myofibroblasts present within the tumour stroma, play a major role in driving tumour progression. We developed a coimplantation tumour xengraft model for experimentally generating CAFs from human mammary fibroblasts. The protocol describes how to establish CAF myofibroblasts that acquire an ability to promote tumourigenesis.
