Agarose Cast

An agarose cast is a solidified agarose gel formed in a mold to create a stable medium for separating and analyzing biological molecules. Agarose is heated in an electrophoresis buffer, poured around a comb to form sample wells, and allowed to cool into a porous matrix; an applied electric field then drives charged nucleic acids through the gel according to size. In immunology and infection research, agarose casts support the analysis of PCR products, immune-receptor sequences, microbial DNA, and other nucleic acid samples. Proper casting produces uniform wells and gel structure, improving resolution, reproducibility, and interpretation of molecular assays.

An indirect additive manufacturing method combining a 3D printing of polymers with a centrifugal casting is outlined for manufacturing 3D octet truss metals (Al and Cu alloys) having a unit cell length of 5 mm with a wall thickness of 0.5 mm.

Mucins are high-molecular-weight glycoconjugates, with size ranging from 0.2 to 200 megadalton (MDa). As a result of their size, mucins do not penetrate conventional polyacrylamide gels and require larger pores for separation. We provide a detailed protocol for mucin agarose gel electrophoresis to assess relative quantification and study polymer assembly.
