Chikungunya Vlps

Chikungunya virus-like particles (VLPs) are noninfectious, genome-free structures that mimic the outer organization of chikungunya virus, providing a platform for studying immune protection. They form when viral structural proteins self-assemble into particles displaying envelope antigens, allowing immune cells to recognize key viral surfaces without viral genome replication. In immunology and infection research, chikungunya VLPs support vaccine development by inducing virus-specific and potentially neutralizing antibody responses while avoiding the risks associated with live-virus systems. They also help researchers investigate antigen presentation, antibody-mediated protection, and immune responses to chikungunya infection, informing safer preventive strategies.

A method to analyze the solvent accessibility of the thiol group of cysteine residues of Maize rayado fino virus (MRFV)-virus-like particles (VLPs) followed by a peptide cross-linking reaction is described. The method takes advantage of the availability of several chemical groups on the surface of the VLPs that can be targets for specific reactions.

Current multiplexed diagnostics to detect Zika, chikungunya, and dengue viruses require complex sample preparation and expensive instrumentation, and are difficult to use in low resource environments. We show a diagnostic that uses isothermal amplification with target-specific strand displaceable probes to detect and differentiate these viruses with high sensitivity and specificity.
