Fluorescent Cell Quantification

Fluorescent cell quantification is a technique for measuring the number, distribution, or signal intensity of labeled cells in a sample, providing quantitative information beyond visual observation. It works by tagging cells or cellular components with fluorescent markers, then detecting emitted light with microscopy or other fluorescence-based imaging systems and analyzing the resulting signals. In immunology and infection research, the method can quantify immune-cell populations, assess pathogen-associated cellular changes, and compare responses across experimental conditions. These measurements support studies of host-pathogen interactions, immune activation, cell recruitment, and treatment effects while enabling reproducible analysis of complex biological samples.

Efferocytosis, the phagocytic removal of apoptotic cells, is required to maintain homeostasis and is facilitated by receptors and signaling pathways that allow for the recognition, engulfment, and internalization of apoptotic cells. Herein, we present a fluorescence microscopy protocol for the quantification of efferocytosis and the activity of efferocytic signaling pathways.

The abundance of neurotransmitter receptors clustered at synapses strongly influences synaptic strength. This method quantifies fluorescently-labeled neurotransmitter receptors in three dimensions with single-synapse resolution in C. elegans, allowing hundreds of synapses to be rapidly characterized within a single sample without distortions introduced by z-plane projection.
