Pad Inhibition

PAD inhibition is the suppression of peptidylarginine deiminase enzymes, which convert protein-bound arginine residues into citrulline and regulate inflammatory immune responses. In infection and immunology, inhibiting PAD4 limits histone citrullination, a process that promotes chromatin decondensation and the formation of neutrophil extracellular traps, or NETs. Researchers use PAD inhibitors to examine how NETosis influences pathogen control, inflammation, and tissue injury, while also assessing whether excessive NET formation contributes to disease. These studies may support the development of targeted strategies that modulate immune activity without broadly suppressing host defense.

Transplantation Into the Mouse Ovarian Fat Pad

We describe an ovarian fad pad transplantation assay suitable for studies of normal and transformed epithelia of the female reproductive tract. The mouse fat pad allows transplantation of large tissue fragments, is easily accessible for surgery and imaging, and provides the most favorable native environment for tissues of Müllerian origin.

This protocol describes the method of determining basal and forskolin-stimulated lipolysis in inguinal fat pads obtained from normal chow diet (NCD) or high fat diet (HFD) ± capsaicin fed wild type mice. As an index for lipolysis, glycerol release was measured from inguinal adipose fat pads.
