Transmission Blocking

Transmission blocking refers to strategies that interrupt the spread of an infectious agent from an infected host to a new host, making it a central goal of infection control and immunology. These approaches work by reducing pathogen replication, shedding, infectivity, or the ability to invade and develop within a recipient or vector; antibodies, vaccines, antimicrobial drugs, and vector-directed interventions can act at these stages. By limiting onward transmission rather than focusing only on symptoms in the infected individual, transmission-blocking measures can reduce outbreak growth, protect vulnerable populations, and support elimination efforts when combined with diagnosis, treatment, and public health surveillance.

Ookluc: A Plasmodium berghei Line for Identifying Transmission-blocking Compounds

2025

Plasmodium sexual development occurs in the mosquito midgut. Targeting the parasite's sexual stages is a promising strategy to block transmission. We developed Ookluc, a transgenic P. berghei expressing nanoluciferase after zygote formation, enabling high-throughput screening of transmission-blocking compounds. This article details the methods for screening using Ookluc.

Sample preparation for cryo-electron microscopy (cryo-EM) is a significant bottleneck in the structure determination workflow of this method. Here, we provide detailed methods for using an easy-to-use, three-dimensionally printed block for the preparation of support films to stabilize samples for transmission EM studies.

Transmission of Multiple Signals through an Optical Fiber Using Wavefront Shaping

We demonstrate the transmission of multiple independent signals through a multimode fiber using wavefront shaping employing a single spatial light modulator. By modulating the wavefront for each signal individually, spatially separated foci are transmitted. Potential applications are multiplexed data transfer in communications engineering and endoscopic light delivery in biophotonics.

Source: Lutz, D., et.al. Assessment of Ultrastructural Neuroplasticity Parameters After In Utero Transduction of the Developing Mouse Brain and Spinal Cord. J. Vis. Exp. (2019).This video demonstrates the preparation and transmission electron microscopy (TEM) analysis of a brain section from a mouse pup. The protocol starts with osmium tetroxide-fixed tissue embedded in resin, followed by precise trimming to isolate the region of interest and ultramicrotomy to produce semithin and ultrathin...

Testing the Physiological Barriers to Viral Transmission in Aphids Using Microinjection

Aphids are effective transmitters of plant viruses. Aphid microinjection of virus, the procedure we will show you today, is a technique allowing researchers to inject virus directly into the hemocoel of the aphid, bypassing the gut, one of the 2 major barriers for virus transmission in a circulative manner. The same technique is also used to inject dsRNA for RNAi.
