Peritoneal Fluid Harvesting

Peritoneal fluid harvesting is a method for collecting fluid from the peritoneal cavity to examine immune cells, soluble mediators, and infectious agents in a localized biological environment. In experimental immunology, sterile fluid is introduced into the cavity and recovered by lavage, allowing resident leukocytes and inflammatory molecules to be sampled when direct aspiration yields little material. The recovered sample can be analyzed by microscopy, cell counting, immunophenotyping, culture, or molecular assays to characterize host responses, pathogen presence, and changes over time. This approach supports studies of innate immunity, peritonitis, host-pathogen interactions, and treatment effects while linking local immune events to disease mechanisms.

In this video, we describe the procedure for isolating low-density neutrophils, or LDNs, from the peritoneal lavage fluid of a postoperative human patient. The LDNs thus obtained can be used in further downstream experiments to understand their role in tumor growth and progression.
