Peroxidase Activity Assay

A peroxidase activity assay is a biochemical method that measures the catalytic activity of peroxidase enzymes, which use hydrogen peroxide to oxidize a suitable electron-donor substrate. In a typical assay, enzyme-driven substrate oxidation produces a colored or luminescent signal, and its intensity is measured to estimate activity under defined reaction conditions. In immunology and infection research, peroxidase assays support enzyme-linked immunosorbent assays, immunohistochemistry, and pathogen detection by revealing antibody or antigen binding through labels such as horseradish peroxidase. Quantifying this signal helps assess immune responses, identify infectious agents, and compare enzyme activity across samples or experimental treatments.

Fast and Specific Assessment of the Halogenating Peroxidase Activity in Leukocyte-enriched Blood Samples

This protocol describes the quick enrichment of leukocytes from small blood samples for a subsequent specific determination of the halogenating peroxidase activity within the cells. The method can be applied to human and non-human material and may contribute to the evaluation of new inflammatory markers.

This paper presents an economical and efficient protocol for examining peroxidase-positive leukocytes in semen. With the assistance of a computer-assisted semen analysis (CASA) system, the concentration of peroxidase-positive leukocytes in semen can be obtained within a total of 60 min, effectively improving the efficiency of andrology laboratory and andrologists.
