Poly(a) Leader Sequence

A poly(A) leader sequence is a stretch of adenosine nucleotides at the 5′ end of certain messenger RNAs, where it can influence transcript stability and protein production. In poxviruses, these leaders are generated during early transcription when the viral RNA polymerase repeatedly slips on a template containing consecutive thymidine residues, adding a variable-length adenosine tract before the coding region. Poly(A) leaders help researchers investigate viral gene expression, translation control, and host–pathogen interactions. Their study also supports the development of improved viral vectors, antiviral strategies, and experimental systems for examining how infections reshape cellular protein synthesis.
