Recombination Efficiency

Recombination efficiency is the extent to which DNA segments are correctly joined during a recombination process, influencing the yield and diversity of the resulting genetic products. In immunology, it describes how effectively developing lymphocytes assemble antibody and T-cell receptor genes through V(D)J recombination, in which RAG1 and RAG2 recognize recombination signal sequences and initiate DNA cleavage before repair by nonhomologous end joining. Efficient recombination supports the generation of diverse antigen receptors and functional B- and T-cell populations. Measuring or manipulating this efficiency helps researchers study immune development, immunodeficiency, infection susceptibility, and strategies for improving engineered immune-cell responses.

Here we describe a protocol based only on cell infection, which improves the efficiency of recombinant parvovirus production by more than 100 fold in comparison to other protocols in use. This protocol relies on the use of a novel adenovirus 5-based helper containing the parvovirus VP transcription unit (Ad-VP).

Plants offer a novel system for the production of pharmaceutical proteins on a commercial scale that is more scalable, cost-efficient and safe than current expression paradigms. In this study, we report a simple and convenient, yet scalable approach to introduce target-gene containing Agrobacterium tumefaciens into plants for protein transient expression.

Homologous recombination techniques greatly advance Drosophila genetics by enabling the creation of molecularly precise mutations. The recent adoption of recombineering allows one to manipulate large pieces of DNA and transform them into Drosophila6. The methods presented here combine these techniques to rapidly generate large homologous recombination vectors.
