Zebrafish Infection

Zebrafish infection refers to the experimental study of host responses to microbial pathogens in Danio rerio, a vertebrate model that supports research in immunology and infection. Researchers introduce bacteria, viruses, fungi, or parasites into embryos, larvae, or adult fish and monitor pathogen growth alongside innate and adaptive immune responses, often using live imaging and genetically modified lines. This model links cellular mechanisms such as inflammation, leukocyte recruitment, and antimicrobial defense with infection outcomes. Its small size, optical transparency during early development, and tractable genetics enable efficient investigation of host-pathogen interactions, antimicrobial therapies, and disease mechanisms relevant to human health.

Transparent zebrafish embryos have proved useful model hosts to visualize and functionally study interactions between innate immune cells and intracellular bacterial pathogens, such as Salmonella typhimurium and Mycobacterium marinum. Micro-injection of bacteria and multi-color fluorescence imaging are essential techniques involved in the application of zebrafish embryo infection models.

Development of a Zebrafish Larval Infection Model Using Microgavage

Source: Li, J., et al. Development of a Larval Zebrafish Infection Model for Clostridioides difficile. J. Vis. Exp. (2020).This video demonstrates the microgavage procedure, where an anesthetized zebrafish larva is immobilized in agarose and orally injected with a pathogenic bacterium using a tracer-guided needle. The non-invasive delivery mimics natural infection, enabling analysis of bacterial replication, toxin release, and intestinal epithelial damage.

(2019).This video demonstrates the method of establishing a foodborne bacterial intestinal infection model in zebrafish larvae by feeding them paramecia infected with fluorescent E. coli.

A Phage Therapy against Pseudomonas aeruginosa Infection in Zebrafish Embryos

2025

The video demonstrates the phage therapy technique targeting Pseudomonas aeruginosa infection in cystic fibrosis zebrafish embryos. Phage therapy effectively diminishes bacterial load and mitigates proinflammatory immune responses in CF zebrafish embryos infected with bacteria, leading to reduced CF embryo lethality.

A Technique to Quantify Fungal Infection in Zebrafish Larvae

2025

This video demonstrates a method to quantify fungal infection in zebrafish larvae. Fungal spores are microinjected into the larval hindbrain ventricle, and the larvae are incubated to allow infection development. Infected larvae are harvested at defined intervals, and infection progress is assessed by enumerating colony-forming units.
