Bile-pancreatic Separation

Bile-pancreatic separation is a surgical principle that routes ingested food separately from bile and pancreatic secretions, altering where digestion and nutrient absorption occur in the intestine. In this configuration, food passes through an alimentary limb while bile and pancreatic enzymes travel through a biliopancreatic limb before mixing with food in a downstream common channel. Used primarily in metabolic and bariatric surgery, the approach can limit nutrient absorption and influence weight and metabolic outcomes. Because reduced digestion and absorption may cause protein, vitamin, and mineral deficiencies, patients require careful nutritional assessment and long-term clinical monitoring.

This protocol explains a method for isolating pancreatic islets by first perfusing collagenase via Ampulla of Vater to digest the exocrine pancreatic tissue and a simplified density gradient centrifugation to purify the islets from mice.
