Gill Stain Analysis

Gill stain analysis is the evaluation and use of Gill’s hematoxylin, a nuclear stain that highlights cellular detail in cytology and histopathology. During staining, hematoxylin is oxidized to hematein and forms a metal-dye complex with an aluminum mordant; this complex binds to acidic components such as nuclear chromatin, producing blue to violet nuclei after bluing. Progressive staining allows color intensity to develop directly in the specimen, while differentiation and controlled timing help limit background staining. In medicine, Gill staining supports assessment of nuclear morphology in tissue sections, cervical smears, and other clinical specimens, improving diagnostic interpretation and providing a basis for monitoring stain quality.
