Perfusate Composition

Perfusate composition is the formulation of a fluid circulated through an isolated organ or tissue to maintain its function, viability, or diagnostic utility outside the body. A perfusate typically balances electrolytes, osmotic agents, buffers, nutrients, and, when needed, oxygen carriers to support cellular metabolism, regulate pH and fluid shifts, and limit ischemic injury during controlled perfusion. In medicine, carefully tailored compositions support organ preservation, ex vivo perfusion, transplantation assessment, and physiological research, while adjustments in temperature, oxygenation, and drug content can help evaluate tissue performance or deliver targeted interventions before clinical use.

Classifying Matter by Composition

2020

Matter: Pure Substances and Mixtures According to its composition, the matter can be classified into two broad categories — pure substances and mixtures. A pure substance is a form of matter that has a constant composition throughout with uniform properties. For example, any sample of sucrose has the same composition and same physical properties, such as melting point, color, and sweetness, regardless of the source from which it is isolated. A mixture is composed of two or more types of...

The porcine model of liver normothermic machine perfusion (NMP), described here, can be successfully used to study NMP as a preservation strategy, a tool for viability assessment, and a platform for organ repair. It holds a high translational value, however it is technically challenging and labor-intensive.

Antegrade Perfusion Model: An Ex Vivo Technique to Perfuse Isolated Mouse Heart

2025

This video describes an antegrade perfusion technique of an isolated mouse heart during which a perfusate is injected through the left ventricle, and eventually, it perfuses the entire myocardium. This method can be used to isolate cardiomyocytes from murine models and generate decellularized hearts for further downstream applications.
