4-hydroxytamoxifen Induction

4-hydroxytamoxifen induction is a chemical method for controlling Cre recombinase activity in genetically modified cells, enabling time-specific gene manipulation in neuroscience. The 4-hydroxytamoxifen metabolite binds the modified estrogen receptor in CreERT2, allowing the fusion protein to enter the nucleus and recombine loxP-flanked DNA sequences. Researchers can therefore activate or delete genes in selected neurons or glial cells at defined developmental stages or after experimental treatment. This temporal and cellular control supports studies of neural circuit formation, synaptic function, behavior, neurodegeneration, and other processes while reducing effects from constitutive gene modification.

Here, we present an overview of the preparation and animal handling procedures required to safely perform medical imaging in an animal biosafety level 4 laboratory. Computed tomography of a mock-infected guinea pig illustrates these procedures that may be used to evaluate the disease caused by a high consequence pathogen.

Here, we present a protocol to induce paralysis and opticospinal inflammation by transfer of aquaporin-4 (AQP4)-specific T cells from AQP4-/- mice into WT mice. In addition, we demonstrate how to use serial optical coherence tomography to monitor visual system dysfunction.

In this video, a genetically modified mouse pup carrying the Ccm2 gene flanked by loxP sites and Cre recombinase fused to an estrogen receptor is used to induce cerebral cavernous malformations (CCMs). The administration of 4-hydroxytamoxifen activates Cre in endothelial cells, leading to the deletion of Ccm2 and the formation of CCMs in the brain.
