Brain Slice Culture

Brain slice culture is an ex vivo method that maintains portions of brain tissue in a controlled laboratory environment, allowing neural circuits to be studied outside the intact organism. Researchers cut brain tissue into thin sections and support the slices in nutrient media under conditions that preserve cellular interactions, synaptic signaling, and aspects of local network activity. This preparation enables direct investigation of neuronal and glial function, synaptic plasticity, development, and responses to drugs or injury. Brain slice culture therefore provides a useful bridge between isolated-cell experiments and whole-animal studies, supporting mechanistic research in neuroscience while reducing the complexity of the intact brain.

This video demonstrates a modified roller tube method for the long-term culturing of brain slices from a rodent model. First, the brain slices are attached to coverslips. Then, flat-sided roller tubes with a hole on the flat side are taken, and the coverslips are positioned on the hole using adhesive discs. Finally, a nutrient medium and a carbon dioxide and air mixture are added to the tube, and the tubes are incubated with a rolling motion.

We describe a method to prepare organotypic hippocampal slices that can be easily adapted to other brain regions. Brain slices are laid on porous membranes and culture media is allowed to form an interface. This method preserves the gross architecture of the hippocampus for up to 2 weeks in culture.
