Dual Reporter Mesc Line

A dual reporter mESC line is a mouse embryonic stem cell line engineered to produce two distinguishable reporter signals, enabling researchers to monitor separate molecular states or cell fates in the same culture. Typically, regulatory DNA sequences linked to cell-type-specific genes drive fluorescent or other detectable reporters, so changes in signal reveal gene activation during self-renewal, differentiation, or lineage progression. In neuroscience, dual reporter lines help track the emergence of neural progenitors and neurons, compare developmental pathways, and evaluate how experimental conditions influence cell fate. They support live-cell imaging, cell sorting, and quantitative analysis of neural differentiation in vitro.

SC1 functions through dual inhibition of Ras- GAP and ERK1. We tested the function of SC1 in supporting mouse ES cell self-renewal in the absence of LIF and showed that SC1 is able to maintain self-renewal of mouse ES cell cultures.

Here, we present a protocol describing a streamlined method for the efficient generation of plasmids expressing both the CRISPR enzyme and associated single guide RNA (sgRNAs). Co-transfection of mammalian cells with this sgRNA/CRISPR vector and a dual luciferase reporter vector that examines double-strand break repair allows evaluation of knockout efficiency.
