Egfp Labeling

EGFP labeling is a fluorescent tagging technique that uses enhanced green fluorescent protein to mark selected cells, proteins, or cellular structures for visualization. In neuroscience, researchers introduce an EGFP-encoding sequence through genetic expression or fuse it to a protein of interest; the resulting protein emits green fluorescence when illuminated, allowing labeled components to be detected by fluorescence microscopy. This approach supports the identification and tracking of neurons, axons, dendrites, and subcellular proteins in cultured cells and nervous tissue. EGFP labeling helps reveal cellular organization, protein localization, and changes in neuronal structure during development, disease, or experimental manipulation.

Here, we describe the methodology used for generating L. panamensis and L. donovani strains expressing the gene for eGFP as a stable integrated transgene using the pLEXSY system. Transfected parasites were cloned by limiting dilution, and clones with the highest fluorescence intensity in both species were selected for further use in drug screening assays.
