Histochemical Analysis

Histochemical analysis is a set of methods that identifies and maps chemical components within cells and tissues, linking molecular composition to tissue structure and function. In practice, tissue sections are treated with selective stains, antibodies, or enzyme-based reagents that bind target molecules or trigger localized reactions, producing visible color or fluorescence for microscopic examination. In neuroscience, these signals help researchers trace neuronal organization, characterize glial and synaptic features, and assess changes associated with development, injury, or disease. By preserving spatial context, histochemical analysis connects molecular observations with neural anatomy and supports comparisons across experimental conditions.

Pre-fixed sections are treated with a straining reagent that highlights active neural clusters, forming dark, high-contrast regions. After mounting and dehydration, cortical modules appear under a bright-field microscope as dark, patchy patterns.

Immunohistochemistry is a powerful lab technique for evaluating protein localization and expression within tissues. Current semi-automated methods for quantitation introduce subjectivity and often create irreproducible results. Herein, we describe methods for multiplexed immunohistochemistry and objective quantitation of protein expression and co-localization using multispectral imaging.

A Neuronal and Astrocyte Co-Culture Assay for High Content Analysis of Neurotoxicity

This article describes a novel protocol and reagent set designed for sensitive measurement of neurotoxic effects of compounds and treatments on co-cultures of neurons and astrocytes using high content analysis. Results demonstrate that high content analysis represents an exciting novel technology for neurotoxicity assessment.
