Longitudinal Recordings

Longitudinal recordings are repeated measurements of the same biological system across multiple time points, allowing researchers to study neural activity and change within an individual rather than comparing separate groups. In neuroscience, researchers use consistent recording methods and behavioral or physiological landmarks to align signals collected over days, weeks, or longer periods, revealing how neural circuits evolve during learning, development, disease, or recovery. This approach preserves individual trajectories, improves sensitivity to gradual or transient changes, and supports analysis of relationships between brain activity and behavior. It is especially valuable for evaluating circuit plasticity, monitoring disease progression, and assessing responses to interventions over time.

Genetically encoded luciferase is a popular non-invasive reporter of gene expression. Use of an automated longitudinal luciferase imaging gas- and temperature-optimized recorder (ALLIGATOR) enables longitudinal recording from bioluminescent cells under a wide range of conditions. Here we show how ALLIGATOR may be used in the context of circadian rhythms research.
