Rod Outer Segment

A rod outer segment is the light-sensitive, cylindrical compartment of a retinal rod photoreceptor that converts photons into neural signals for vision in dim conditions. It contains stacked membranous discs enriched with rhodopsin and other phototransduction proteins; when light activates rhodopsin, a G protein signaling cascade lowers cyclic GMP, closes ion channels, and hyperpolarizes the cell. Rod outer segments are continually renewed as new discs form near the base and older material is shed at the tip. Their structure and function are central to studying retinal physiology, inherited blindness, photoreceptor degeneration, and potential vision-restoring therapies.

Outer-Boundary Assisted Segmentation and Quantification of Trabecular Bones by an Imagej Plugin

We present a workflow for segmenting and quantifying trabecular bones for 2D and 3D images based on the bone's outer boundary using an ImageJ plugin. This approach is more efficient and accurate than the current manual hand-contouring approach, and provides layer-by-layer quantifications, which are not available in current commercial software.

This article describes the protocol for the purification of photoreceptor outer segment fragments (POS) via ultracentrifugation from porcine/bovine retinae using homogenization and sucrose gradient centrifugation. This protocol allows the preparation of large stocks of POS aliquots, labeled or unlabeled, that can then be stored at -80 °C.
