Tumor Tissue Processing

Tumor tissue processing is the preparation of tumor specimens for microscopic, molecular, and pathological analysis, preserving cellular architecture and disease-related information. The workflow typically involves collecting and dissecting tissue, stabilizing it through fixation or freezing, embedding it in a supporting medium, and cutting thin sections for staining or imaging. In neuroscience, careful processing of brain and nervous system tumors helps reveal interactions between malignant cells, neurons, glial cells, blood vessels, and surrounding tissue. These preparations support tumor classification, biomarker assessment, research on invasion and treatment response, and the development of models for investigating cancer biology in the nervous system.

Tissue Engineering of a Human 3D in vitro Tumor Test System

Methods to create human 3D tumor tissues as test systems are described. These technologies are based on a decellularized Biological Vascularized Scaffold (BioVaSc), primary human cells and a tumor cell line, which can be cultured under static as well as under dynamic conditions in a flow bioreactor.

The identification of brain tumor initiating cells (BTICs), the rare cells within a heterogeneous tumor possessing stem cell properties, provides new insights into human brain tumor pathogenesis. We have refined specific culture conditions to enrich for BTICs, and we routinely use flow cytometry to further enrich these populations. Self-renewal assays and transcript analysis by single cell RT-PCR can subsequently be performed on these isolated cells.
