Hydrophilic Drugs

Hydrophilic drugs are water-soluble medicines whose chemical properties influence how they dissolve, move through body fluids, cross biological barriers, and reach their targets. Because they have limited ability to partition into lipid membranes, their absorption and distribution often depend on aqueous channels, paracellular pathways, membrane transporters, or administration routes that bypass gastrointestinal barriers. These characteristics affect oral bioavailability, tissue penetration, onset and duration of action, and renal elimination. Understanding hydrophilic drug behavior helps pharmacologists select suitable formulations, predict drug concentrations, assess transporter-mediated interactions, and optimize therapies for infections, cancer, cardiovascular disease, and other conditions.

This article presents a protocol for the production of protein-based nanoparticles that changes the hydrophobic surface to hydrophilic. The produced nanoparticle is an assembly of gliadin-cyanoacrylate diblock copolymers. Spray coating with the produced nanoparticle changes the surface of target material to a hydrophilic surface.

Synthesis of Poly(N-isopropylacrylamide) Janus Microhydrogels for Anisotropic Thermo-responsiveness and Organophilic/Hydrophilic Loading Capability

We present a protocol to synthesize Janus microhydrogels composed entirely of the same base material, poly(N-isopropylacrylamide) (PNIPAAm), with a clearly compartmentalized structure base on the phase separation of a supersaturated NIPAAm monomer solution. The synthesized Janus microhydrogels show unique properties such as anisotropic thermo-responsiveness and organophilic/hydrophilic loading capability.

Preparation and Characterization of Nanoliposomes for the Entrapment of Bioactive Hydrophilic Globular Proteins

This study describes classical hydration using the thin lipid film method for nanoliposome preparation followed by nanoparticle characterization. A 47 kDa-hydrophilic and globular protein, tarin, is successfully encapsulated as a strategy to improve stability, avoid fast clearance, and promote controlled release. The method can be adapted to hydrophobic molecules encapsulation.
