Muscle Tone Reduction

Muscle tone reduction is the pharmacological lowering of involuntary resting muscle activity and resistance to movement, an important approach for managing spasticity and muscle hyperactivity. Drugs achieve this by decreasing excitatory signaling in the central nervous system or at the neuromuscular junction; for example, baclofen activates GABA-B receptors, tizanidine stimulates alpha-2 adrenergic receptors, and botulinum toxin inhibits acetylcholine release locally. These mechanisms can improve mobility, comfort, posture, and rehabilitation outcomes in conditions involving abnormal muscle tone. Because excessive reduction may cause weakness, sedation, or impaired function, treatment requires careful selection, dosing, and monitoring.

We have developed a device (Twister) to study the regulation of tonic muscle activity during active postural maintenance. Twister measures torsional resistance and muscular responses in standing subjects during twisting of the body axis. The device can be flexibly configured to study various aspects of tonic control across the neck, trunk, and/or hips.
