Two Types of Assays for Detecting Frog Sperm Chemoattraction

在无脊椎动物中，精子的化学趋向性可能足够显著，以至于可以将含有趋化肽的移液管插入精子悬液中，通过显微镜直接观察到精子在移液管周围的聚集现象1。而在脊椎动物（如青蛙、啮齿类动物和人类）中，精子化学趋向性的检测更为困难，需要采用定量分析方法。这类分析方法主要分为两类：一类是量化精子向趋化因子源移动情况的实验，即所谓的精子聚集实验；另一类则是实际追踪单个精子游动轨迹的实验。
精子聚集实验相对快速，可在一天内完成数十甚至数百次实验，从而能够较快地开展剂量反应曲线和时间过程的研究。这类实验已被广泛用于表征许多已建立的化学趋化系统——例如，中性粒细胞对细菌肽类的趋化作用以及精子对卵泡液的趋化作用。精子追踪实验则更为耗时，但能提供化学趋化物如何实际改变精子游动路径的额外数据。此类实验对于证明精子运动方向相对于化学趋化梯度轴的定向性，以及观察使精子更接近卵子的特征性转向或方向变化至关重要。
本文描述了上述两种类型实验所使用的方法。其中精子趋化聚集实验采用的是“双室”实验法。将两栖动物的精子置于带有聚碳酸酯滤膜底部的组织培养板小室中，滤膜孔径为12 μm。将含有精子的小室插入含有缓冲液的组织培养板孔中，并用移液器将趋化因子小心加入底部孔中，使其位于滤膜底部与孔壁交界处（见图1）。孵育一段时间后，小心移除上层含有精子储存液的小室，收集穿过滤膜进入下室的精子，经离心形成沉淀后，使用血细胞计数板或流式细胞仪进行计数。
精子追踪实验采用Zigmond小室，该装置最初用于观察中性粒细胞趋化性，后由Giojalas及其同事加以改进，用于精子观察2,3。该小室由一块厚玻璃载片制成，其上加工出两条垂直的槽，中间由一条宽1 mm的观察平台隔开。盖上盖玻片后，将精子加入其中一条槽，化学趋化剂加入另一条槽，通过视频显微镜观察单个精子的运动。随后利用软件分析视频影像，获取精子在x-y平面内随时间变化的二维细胞运动轨迹（即xyt数据集），从而形成每条精子的运动轨迹。

本文介绍了用于双室检测的两种不同精子趋化性评估方法。首先，将激活的蛙类精子加入具有多孔膜底部的插入式培养小室中，并将小室置于24孔板的孔内。待精子就位后，小心地向孔的底部加入趋化因子，以建立趋化性梯度。接下来，将精子进行孵育，以使细胞有足够时间在化学趋化梯度作用下穿过微孔膜。最后，移除插入式培养插件，利用血细胞计数板对在双室实验中迁移至下室底部的精子进行计数。最终，可通过例如Zigman chamber示踪实验中的剂量-反应曲线分析，对精子趋化因子的活性进行比较和定量。首先将盖玻片用硅油固定在Zigman小室上，并倒置覆盖在显微镜的圆形开口上。然后向小室的一侧加入活化的青蛙精子。接着，向另一侧加入趋化吸引剂溶液。五分钟后，化学趋向性梯度形成，此时可对精子沿梯度游动的过程进行录像。最终，可通过分析齐格蒙德小室追踪实验所获得的影像，确定精子的运动速度以及精子在化学趋化剂梯度作用下的趋化运动模式和运动轨迹。这两种检测精子趋化性的方法互为补充，可产生不同类型的实验数据。双室实验允许精子穿过微孔膜，从而定量测定精子对化学趋化剂的反应。双室实验的可视化演示至关重要，因为在实验开始时添加精子和化学趋化剂，以及在实验结束时移除精子和插入物的步骤，均需要微量移液器操作的精确定位。Zigmond chamber 实验利用视频记录精子在化学趋化梯度中的游动行为。这可以确定速度和方向等动态参数。与双室实验相比，泽格蒙德小室追踪实验的主要优势在于能够对精子游动进行可视化，并利用多个动态参数进行分析。通过在睾丸一端打孔，并在另一端使用注射器和针头注入缓冲液，将精子从蛙的睾丸中冲出。从蛙类睾丸中冲洗出精子后，通过血细胞计数板估算所收集的细胞数量。然后用卵母细胞林格液将原液精子悬液稀释，使精子密度达到每毫升两千万个细胞（2×10⁷ cells/mL）。通过将5 µL精子与F1缓冲液按1:10比例稀释，并使用相差光学显微镜观察精子运动情况，以评估精子活力。接下来，用微量移液器将700微升F1缓冲液加入塑料24孔组织培养板的每个孔中。然后，在室温下，将100微升精子悬液与900微升F1缓冲液在微型离心管中混合，通过渗透冲击激活每次检测所需的精子活力。将一个直径12毫米、孔径12微米的插入式培养小室放入盛有缓冲液的孔中，确保小室偏离中心放置，一侧留出空隙。然后使用带有切口吸头的微量移液器，立即将400 µL运动激活的精子转移至小室中，将精子悬液沿插入式小室的壁加入，使其自然流下并接触插入式小室底部的滤膜。接着，小心地用微量移液器将50 µL趋化吸引剂加入到每孔板中，加样位置为小室与偏心插入式小室之间的空隙处。注意将液滴加在孔的侧壁与底部交界处。移液后避免对体系造成扰动。待所有检测设置完成后。在第一个检测开始孵育50分钟后，将培养板置于室温下。随后，用一只手小心地固定插入物。通过移液器移除上室中大部分或全部精子悬液，以终止检测。将精子从上层小室转移后，立即取出插入物并弃去。随后混匀每个孔中的精子悬液，以重悬沉降到孔底的任何精子。然后将每个孔中的全部精子悬液转移至各自的微量离心管中。加入甲醛，使其终浓度为0.5%（体积比）。将每管中固定的精子进行沉淀。使用10秒点动按钮，在小型微量离心机上离心。然后从每支试管中除去上清液，仅保留 100 微升，并用该体积重新悬浮沉淀。接着，取 20 微升精子悬液，用蒸馏水按 1:10 比例稀释，并在血细胞计数板上计数精子数量。使用直立显微镜的 40 × 物镜，双室检测可提供适用于剂量反应研究或不同趋化物之间比较的定量结果。例如，蛙卵水中含有精子化学趋化剂，即图中红色符号所示的诱引素。随着精子化学趋化剂剂量的增加，穿过微孔膜的精子数量先升高后降低。相比之下，在相同剂量范围内，由白色符号代表的牛血清白蛋白几乎无影响。开始Zigman小室实验前，首先确认倒置显微镜工作站已准备就绪，可用于录像。安装显微镜，并启动相机控制软件，以每秒七帧的速度采集图像。检查摄像头是否已对准观察平台，并确认输出信号已传输至计算机显示器。接着，检查显微镜视野是否包含观察平台宽度的大部分或全部区域。然后，用卵母细胞任氏液制备青蛙精子悬液，并用F1缓冲液将趋化引诱物稀释至适当浓度，两者均置于冰上保存，待用。然后从一个洁净、干燥的Zigmund小室开始。使用微量移液器在距离每个trth外缘约五毫米处，平行于外缘的位置，滴加一条约四微升的硅油。接着将一块22乘40毫米的盖玻片放置于小室上，使硅油均匀扩散至每个trth的外缘。现在将腔室倒置，并将其放置在显微镜载物台的大圆形切口上方。注意避免盖玻片与载物台接触，通过在室温下将xap精子与F1缓冲液按1:10比例混合，用20微升的卵母细胞林格氏缓冲液激活精子。使用带有切口吸头的微量移液器，立即吸取70微升已激活运动能力的精子。然后以较低角度持握微量移液器，将吸头置于侧面开口处，通过毛细作用填充trth和桥接部分。接着使用趋化引诱剂溶液以同样方式填充另一侧的trth，因为xap精子的运动寿命有限。在三分钟内开始录像，并持续五分钟。Zigman小室追踪实验可实现对精子在化学趋化剂梯度中游动的视频显微观察。本视频展示了精子在观察平台上游动的记录，其运动轨迹如图中所示，以红色线条标出。精子轨迹由多个片段组成，每个片段用箭头标示，代表精子在连续视频帧之间的运动路径。这些轨迹片段描绘了单个精子的游动行为，并突显出诸如环形运动和转向等模式。根据这些数据，可以确定每个片段的速度和方向，或分析单条轨迹或一组轨迹中所有片段的特性。例如，净线性距离。连接轨迹起点与终点的矢量在 x 轴方向上具有分量，并且与 x 轴成一定角度 θ，如图中红色所示。当存在趋化因子浓度梯度时，θ 角应减小，而沿 x 轴方向的运动距离应增加。这些变化表明精子倾向于向上游趋化因子浓度梯度方向游动。在进行上述两种实验时，必须注意在每个装置中正确加载精子和趋化因子，这是获得可重复的精子趋化梯度响应结果的关键。
