1. 仪器、培养基和培养皿的准备

3. 肠囊制备
<ol>
	<li>准备一支1 ml注射器，内含300 µl待测化合物或分子。为评估黏膜完整性，可使用浓度为1 mg/ml、分子量为4,400的FITC-葡聚糖溶液。为提高检测灵敏度，也可使用分子量范围在4,400–70,000 Da之间的探针。将小型动物血管导管牢固地安装在注射器上。</li>
	<li>从肠段开口处量取5 cm，用缝合线环在此位置将肠段牢固结扎关闭。在肠管开口处轻轻套上预先打结的缝合线环，插入钝头导管。拉紧线环以固定肠段，并从注射器向肠腔内注入300 µl溶液，确保全部液体完全注入。</li>
	<li>在撤出导管的同时，同步收紧缝合线环，以确保肠囊闭合严密。将肠囊从肠管上剪下，放入盛有20 ml已预热至37 °C的充氧培养基的50 ml锥形管中。</li>
</ol>
4. 通透性测定
<ol>
	<li>将装有肠段的锥形管置于设定为37 °C的恒温水浴中。在0、30、60、90和120分钟的时间点，从锥形管中取100 µl样品并转移至96孔板中，每次取样后立即补充100 µl新鲜培养基。</li>
	<li>最后一次取样后，沿缝合处及肠段长度方向剪开肠段，暴露黏膜表面。</li>
	<li>测量每段肠组织的长度和宽度。如需要，可将肠段快速冷冻后于-80 °C保存，用于蛋白质或生化分析；或者 alternatively，将其保存于RNA稳定溶液中用于分子检测。</li>
	<li>构建FITC标记分子的标准曲线，稀释梯度为1至1 × 10-6的对数稀释系列。</li>
	<li>使用荧光酶标仪检测样品及标准品的FITC信号，FITC激发/发射波长为495 nm/519 nm。</li>
